Product Overview:

BIOPHEN™ CS-11(22) Factor Xa Chromogenic Substrate is a synthetic chromogenic substrate designed for use in research applications requiring the measurement of Factor Xa activity. Research use only reagent for professional use only.

Principle and Application:

This synthetic substrate is cleaved by Factor Xa, releasing the chromogenic group p-Nitroaniline (pNA). The rate of pNA release, measured photometrically, is directly proportional to the Factor Xa activity. The substrate is a mixture of two peptide sequences: Bz-Ile-Glu(γ-OCH₃)-Gly-Arg-pNA and Bz-Ile-Glu(γ-OH)-Gly-Arg-pNA. It is intended for all research studies and laboratory protocols where a source of chromogenic substrate for Factor Xa is required.

Product Compositionand Specifications:

The product is supplied as a single vial containing 25 mg of lyophilized BIOPHEN™ CS-11(22) with Mannitol as a bulking agent. The substrate is a highly purified and stabilized synthetic peptide.

• Peptide Sequence: Mixture (50%-50%) of Bz-Ile-Glu(γ-OCH₃)-Gly-Arg-pNA (form 1) and Bz-Ile-Glu(γ-OH)Gly-Arg-pNA (form 2)
• Molecular Weight: 711.8 (form 1) and 697.7 (form 2)
• Purity Grade: > 95%
• pNA Content: < 0.1% (as measured by RP-HPLC)

Performance Characteristics:

The substrate demonstrates high specificity for Factor Xa relative to other serine proteases.

Note: Assay conditions must be duly established for rendering the assay conditions totally specific for Factor Xa when this substrate is used.

Storage and Stability:

• Unopened (Lyophilized) Vial: Store at 2-8°C, protected from light. The product is stable until the expiration date printed on the label.
• Reconstituted Substrate:
  • Stable for 3 months at 2-8°C.
  • Stable for 7 days at room temperature.
  • Important: Do not freeze the reconstituted solution.
